2. ARMS2 / HTRA1
Located on chromosome 10, in 2025, the dominant genes influencing AMD at this position were confirmed by Nature Communications to be HTRA1 and its related non-coding RNA, HTRA1-AS1. For an individual carrying the CFH and ARMS2/HTRA1 risk variants, the risk of AMD could be increased by as much as 106-fold.

It may seem to-, but it is not known what pattern of skipping, if any, exists.AMD is multifactorial; many genes work together, along with your lifestyle. Between 15% to 20% of patients with AMD have one first-degree relative, and an equal number or more have no family history whatsoever.

Take AREDS2 supplements. The tested vitamin-and-mineral formula (vitamins C and E, zinc, copper, lutein, and zeaxanthin) has been shown to slow the progression of dry-to-wet AMD. Consult with your doctor first.

Get moving! Regular exercise, a healthy body weight, and well-managed blood pressure are all good for your retinas.

Have regular eye exams. Early AMD often has no visible signs or symptoms. A dilated eye exam can detect drusen before the disease causes significant vision loss. Schedule annual exams if macular degeneration is in your family history.
Should You Get Genetic Testing?
The American Academy of Ophthalmology currently recommends against genetic testing for AMD on a routine basis. Knowing your gene variants does not alter routine clinical management at this point.
There may be a benefit to testing if you are involved in a clinical trial, have a significant family history, or are seeking risk stratification by a retina specialist. Before pursuing commercial AMD testing, speak with a genetic counselor.
What’s New in AMD Research (2025–2026)
The December 2005 publication in Nature Communications pinpointed HTRA1 and its long noncoding RNA, HTRA1-AS1, as master regulators at the gene locus of chromosome 10. Also, it revealed a possible new pathway for gene therapy. Gene therapy is an area of research that is heavily studied through clinical trials and may prevent AMD in the first place.
